总咖啡酸酯对IgA肾病大鼠肾脏系膜CD71TGF-β1表达的影响

         

摘要

Objective: To explore the therapeutic effects and mechanism of total caffeoyl quinic acid ester(TCaf) on the expression of CD71 and TGF-β1 in mesangial area of Sprague Dawley(SD) rats with experimental IgA nephropathy. Method: IgA nephropathy model had been experimentally induced in SD rats of model group by oral administration of bovine serum albumin, followed by staphylococcal enterotoxin B ,freund's adjuvant and carbon albumin, TCaf Stoup rats had been intraperitoneally administered with TCaf(3mg/kg per day). ImmUnohistochemistry and the RT-PCR methods were used to respectively examine the expression of the protein and mRNA of CDT1 and TGF-β1 in the renal tissue. Result: The administration of TCaf to rats with IgA nephropathy significantly decreased the levels of 24-hrs urinary protein excretion, serum ereatinine and blood urea nitrogen in comparison with those of non-treated rat( P<0.05). Expression of CD71 and TGF-β1 in glomeruli of experimental IgA nephropathy decreased respectively after treatment with TCaf(P <0.05 ). Conclusion: TCaf could effectively reduce hematuria, proteinuria. TCaf could also remarkably inhibit the expression of CDT1 and TGF-β1 in mesangial area of IgA nephropathy in experimental rats.%目的:探讨总咖啡酸酯(Total Caffeoylquinic Acid Ester,TCaf)对IgA肾病(IgAN)大鼠肾脏系膜CD71与TGF-β<,1>,表达的影响.方法:建立IgAN大鼠模型,随机分成模型组(IgAN组)、总咖啡酸酯组(TCaf组)和对照组.用免疫组化和逆转录-聚合酶链反应(RT-PCR)方法分别检测肾组织CD71和TGF-β<,1>蛋白和基因表达水平.结果:与IgAN组比较,TCaf组大鼠24h尿蛋白定量减少、血清肌酐和尿素氮浓度降低;TCaf组在基因和蛋白水平均能够有效抑制CD71和TGF-β<,1>在肾组织中的表达.结论:总咖啡酸酯能改善IgAN大鼠的血尿和蛋白尿,其下调CD71、TGF-β<,1>在肾脏的表达,可能是总咖啡酸酯延缓肾脏纤维化的机制之一.
