总咖啡酸酯对IgA肾病大鼠模型作用的实验研究

             

摘要

To study the influences of the total coffeic ester (TCaf) on urine and kidney pathology in experimental rats with IgA nephropathy (IgAN). Methods 36 male SD mrs were randomly divided into IgAN model group, the total coffeic ester group (TCaf group) and the control group after the IgAN model rots were successfully established by mucous membrane irnmunifaction. Urinary protein, urine red blood cell count, blood serum creatinine, blood urea nitrogen were detected. Immune complex deposition in the kidney and mesangial matrix hyperplasia was also observed by immunofluorescence and light microscopy. A semiquantitafive score was used to evaluate the degree of renal pathologic lesions. Results Compared with model group, ester group of the total 24-hour coffee urine protein, urine red blood cell count, blood serum creatinine, blood urea nitrogen were significantly improved, and immune complex deposition in the kidney significantly reduced, the scores of renal pathologic lesions were significantly reduced. Conclusion The total Caffeic acid esters can improve hematuria and proteinuria, lower serum urea nitrogen and creatinine, reduce immune complex deposition in kidney tissue, and reduce proliferation of mesangial matrix of IgAN rats.%目的 研究总咖啡酸酯(TCaf)对实验性IgA肾病(IgAN)大鼠的尿液及肾脏病理的影响.方法 采用黏模免疫法建立IgAN大鼠模型并验证成功后,将36只雄性SD大白鼠随机分成IgAN模型组、总咖啡酸酯组(TCaf组)、对照组.检测尿蛋白定量、尿红细胞计数、血清肌酐、尿素氮,免疫荧光和光镜观察免疫复合物在肾脏的沉积及系膜区基质增生程度;采用半定量病理分析系统分析肾小球和肾小管损坏程度.结果 与模型组比较,总咖啡酸酯组24 h尿蛋白定量、尿红细胞计数、血清肌酐、尿素氮有明显改善,且免疫复合物在肾脏的沉积明显减少,病理评分明显降低.结论 总咖啡酸酯能改善IgAN模型大鼠的血尿和蛋白尿,降低血清尿素氮和肌酐,减轻免疫复合物在肾组织中的沉积、减轻系膜区基质增生,从而保护肾脏.
