目的:探讨干扰素诱导跨膜蛋白抑制小儿流感病毒的作用及其机制。方法采用实时荧光定量技术对基因表达进行定量分析,先把基因表达进行归一化,然后对实时荧光定量技术的检测条件进行调整,分析蛋白基因扩增的稳定性、灵敏性以及特异性,建立一个体系方法对内参基因、目的基因在细胞中的表达水平进行准确的测定,分析流感病毒NS蛋白和IFITM123表达水平之间的关系,并对IFITM123的蛋白表达量进行定量检测。结果 A549细胞株经过干扰素的诱导后,IFITM1、IF-ITM2、IFITM3的DNA拷贝数都明显增加。结论 IFITM1、IFITM2、IFITM3对于流感病毒NS蛋白具有抑制的作用。%Objective To study the interferon induced transmembrane protein to inhibit the action of the pediatric influenza virus and its mechanism. Methods Real time fluorescence quantitative technique carries on the quantitative analysis of gene expression, gene expression of normalized first, and then to adjust the technology of real-time fluorescence quantitative detection conditions, anal-ysis of the stability of the protein gene amplification, sensitivity and specificity, and so on, to build a system method for internal genes, purpose, accurately determine the level of gene expression in the cell, the analysis of the influenza virus and NS protein IF-ITM1/2/3, the relationship between the expression level of and the IFITM1/2/3 of the quantity of protein expression was quantitative detection. Results The A549 cell lines after interferon inducing IFITM1, IFITM2, IFITM3 DNA copy number increased significant-ly. Conclusion IFITM1, IFITM2, IFITM3 NS for influenza virus protein have inhibition effects.
