microRNA-34a对人脑胶质瘤细胞生物学特性的影响

             

摘要

Objective To investigate the effect of microRNA-34a on the biological characteristics of human brain glioma cells. Methods A total of 100 patients with glioma who were admitted in Department of Neurosurgery, Affiliated Hospital of North Sichuan Medical College from March 2015 to March 2016 were enrolled as the subjects. The glioma tissues were collected. Forty patients with brain trauma treated by decompression in our hospital were select-ed as controls, and normal brain tissue samples were collected during the operation. The expression levels of microR-NA-34a in glioma tissues and normal brain tissues were detected by real-time quantitative PCR. The human cerebral gli-oma cell line U87 after transfection were divided into the blank control group, nonsense sequence transfection group and microRNA-34a transfection group. The effect of microRNA-34a on the proliferation, apoptosis, migration and invasion ability of human glioma cell line U87 was detected. Results The relative expression level of microRNA-34a in glioma tissues was significantly lower than that in normal brain tissues, (0.53±0.48) vs (1.38±0.25), P<0.05. The relative expres-sion level of microRNA-34a showed significantly differences in different deteriorated glioma tissues (P<0.05). 48 hoursafter transfection, the cell growth inhibition rate and cell apoptosis rate after transfection in microRNA-34a transfection group were significantly higher than those in the blank control group and the nonsense sequence transfection group (P<0.05). The number of migrating cells in 24 hours after scratch and number of cells penetrating membrane in 12 hours were significantly lower than those in the blank control group and nonsense sequence transfection group (P<0.05). Conclusion microRNA-34a can inhibit the proliferation, migration and invasion of glioma cells and promote the apop-tosis of tumor cells.%目的 研究microRNA-34a对人脑胶质瘤细胞生物学特性的影响.方法 选取南充市川北医学院附属医院2015年3月至2016年3月收治的100例胶质瘤患者为研究对象,采集胶质瘤组织样本,选取同期在我院接受减压术治疗的40例脑外伤患者为对照,术中采集正常脑组织样本.采用实时荧光定量PCR检测microRNA-34a在胶质瘤组织与正常脑组织样本中表达水平.将转染后人脑胶质瘤细胞系U87分为空白对照组、无义序列转染组及microRNA-34a转染组,检测microRNA-34a对人脑胶质瘤细胞系U87增殖、凋亡、迁移、侵袭能力的影响.结果 胶质瘤组织microRNA-34a相对表达量为(0.53±0.48),显著低于正常脑组织的(1.38±0.25),差异有统计学意义(P<0.05).不同恶化程度胶质瘤组织microRNA-34a相对表达量差异有统计学意义(P<0.05).microRNA-34a转染组转染后48 h细胞生长抑制率、转染后细胞凋亡率显著高于空白对照组与无义序列转染组,划痕24 h后细胞迁移数、细胞12 h穿膜数显著低于空白对照组与无义序列转染组,差异均有统计学意义(P<0.05).结论 microRNA-34a具有抑制胶质瘤细胞增殖、迁移、侵袭,促进肿瘤细胞凋亡等作用.
