高效液相色谱法测定不同产地地骨皮中东莨菪内酯的含量

摘要

A method for determination of scopoletin in Lycii Cortex by High performance liquid chromatography was devel⁃oped. Scopoletin was separated on a ZORBAX C18 chromatographic column with 40% methanol(including 0.2% phosphor⁃ic acid)as mobile phase,detection wavelength was 345nm. The calibration curve of scopoletin showed good linearity in the range of 15~0.12μg/mL,the correlation coefficients(R)was 0.9996. The detection limit was 0.067μg/mL. The method was simply,accurate and showed good repeatability and high sensitivity,it can be used for the determination of scopoletin in Lycii Cortex from different areas.%建立高效液相色谱测定地骨皮中东莨菪内酯含量的方法。方法采用ZORBAX C18色谱柱，以40%甲醇水溶液（含0.2%磷酸）为流动相，检测波长为345nm。东莨菪内酯在15~0.12μg/mL 的范围内呈良好的线性关系（r=0.9996），检测限为0.067μg/mL。该方法操作简单、灵敏度高、精密度良好，可准确测定地骨皮中东莨菪内酯的含量。利用建立的方法测定了国内不同产地地骨皮中东莨菪内酯的含量。
