强心液对心衰大鼠心功能改善作用的实验研究

     

摘要

Objective:To observe the effect of Qiangxin Solution on cardiac function in rats with heart failure (HF) and explore its mechanism.Methods:HF models were induced by adriamycin,and were randomly divided into model group,Qiangxin Solution high dose group,low dose group and Captopril group,and the normal control group was established.Continuous filling of stomach for 4 weeks.The cardiac function of rats was evaluated by echocardiography,and the levels of aldosterone (ALD),atrial natriuretic peptide (ANP) and angiotensin Ⅱ (AngⅡ) in plasma were measured by radioimmunoassay (RIA).Results:Compared with the normal control group,left ventricular end diastolic diameter (LVEDd) and left ventricular end systolic diameter (LV-EDs) of model group increased significantly (P < 0.05),ejection fraction (EF) decreased significantly (P <0.05);EF of Qiangxin Solution high and low dose group were raised and LVEDd,LVEDs were shortened significantly.PlasmALD,Ang Ⅱ and ANP levels in model group rats were increased significantly to compare with normal(P <0.05),these indexes in Qiangxin Solution high and low dose group and Captopril group were lowered significantly to compare with model group(P < 0.05).Conclusion:Qiangxin Solution can improve cardiac function of HF rats and its mechanism may be related to decrease plasma renin angiotensin aldosterone system (RAAS) intensified level especially in the ALD and AnglⅡ level,improve the cardiac afterload.%目的:观察强心液对心力衰竭(HF)大鼠心功能的影响并探讨其作用机制.方法:采用阿霉素诱导法复制HF大鼠模型,随机分为模型组、强心液高、低剂量组和卡托普利组,同时设立正常对照组,连续灌胃给药4周.心脏超声评价各组大鼠的心功能,放射免疫法测定各组大鼠血浆中的醛固酮(ALD)、心钠肽(ANP)和血管紧张素Ⅱ(AngⅡ)含量.结果:与正常对照组比较,模型组左室舒张末内径(LVEDd)和左室收缩末内径(LVEDs)明显增大(P<0.05),射血分数(EF)明显下降(P<0.05);强心液高、低剂量组均能提高HF大鼠的EF,同时强心液能有效改善大鼠心功能并缩短LVEDd和LVEDs.模型组大鼠血浆ALD、ANP和AngⅡ水平明显高于正常对照组(P<0.05);强心液高、低剂量组和卡托普利片组均能下调HF大鼠血浆中的ALD、ANP及AngⅡ的浓度(P<0.05).结论:强心液能缩短HF大鼠的LVEDd和LV-EDs,逆转心肌重构,从而改善心功能,其机制可能与降低HF大鼠血浆肾素-血管紧张素-醛固酮系统(RAAS)的激化水平特别是ALD和AnglI水平,改善心脏后负荷有关.
