乌头碱对心衰细胞强心作用的量-时-效关系的实验研究

摘要

Objective:To observe the dose-time-effect relationship in cardiac effect of heart failure cells induced by H2O2. Methods: I'hrae heart failure models,establised by H;O; and myocardial cells of neonatal rats,were analyzed by the uniform design method. These models were divided into several groups which were used respectively by the dose of 5 |Amol/L,10 u.mol/L,25 p.mol/L,50 |unol/L and 100 (Amol/L for 0.5 h.,1 h,2 h,4 h and 8 h sepa rately. ResultS:The best predicted combination of uniform design was the 50 (Amol/L of aconitine effect for 0.5 h. Meanwhile,the validation experiments also confirmed that. Conclusion:The cardiac effect by aconitine on a cell model of heart failure is significant. The best combination of dose-time is 50 (imol / L and 0.5 h.%目的 观察乌头碱对H2O2构建的心衰细胞模型的强心作用的量-时-效关系.方法 利用原代培养的乳鼠心肌细胞,加H2O2构建心衰模型,采用均匀设计,分别加5、10、25、50、100 μmol/L的乌头碱作用0.5、1、2、4h及8h后测MTT,然后进行验证实验.结果 均匀设计预测的最佳组合为乌头碱50 μmol/L作用0.5 h,验证实验也证实如此.结论 乌头碱对心衰模型有明显强心作用,其最佳量-时-效组合为50 μmol/L的乌头碱作用与心衰模型0.5h.
