桃核承气汤对大鼠糖尿病大血管Toll样受体表达的影响

         

摘要

目的:观测桃核承气汤早期干预对糖尿病鼠大血管病变中TOLL样受体2(TLR-2)、TOLL样受体4(TLR-4)及转化生长因子(TGF-β)、胰岛素样生长因子1(IGF-1)表达的影响.方法:雄性SD大鼠130只,正常对照组(A组)30只,其余100只制造糖尿病模型,分为模型对照组(B组)、中药早期干预组(C组)及中药治疗组(D组).A组不予药物干预;B组给予0.9%氯化钠10 mL·Kg-1·d-1灌胃;C组自造模前予桃核承气汤水溶液灌胃0.9 g·Kg-1·d-1,造模后继续给予桃核承气汤0.9 g·Kg-1·d-1灌胃;D组造模后给予桃核承气汤0.9 g·Kg-1·d-1灌胃.药物干预第1、12、20周处死,分别取材.免疫组化法检测糖尿病大鼠大血管中TLR-2、TLR-4、TGF-β、IGF-1表达情况.结果:药物干预20周后C组大鼠股动脉TLR-2为(3.750±1.282) pg/mL,较B组[(9.333±2.338) pg/mL]明显降低(p<0.05),D组与B组大鼠股动脉TLR-2表达无明显差异(P>0.05).C组及D组大鼠股动脉TLR-4表达为(3.750±1.282)pg/mL、(5.750±1.982) pg/mL,较B组[(10.333±2.658) pg/mL]明显降低(P<0.05),且C组对大鼠股动脉TLR-4表达干预作用强于D组(P<0.05).随着用药时间延长,C组与D组大鼠股动脉TLR-2、TLR-4表达干预作用趋于相近(P>0.05).大鼠股动脉TGF-β C组为(403.438± 11.273) pmol/L,D组为(535.313±12.384) pmol/L,均明显降低(P<0.05);IGF-1 C组为(1104.375±55.495) pmol/L,D组为(861.000±28.656),均有明显升高(P<0.05),且C组作用较D组强.结论:中药桃仁承气汤可减轻糖尿病大血管纤维化,且早期干预对糖尿病大血管纤维化有预防作用.%Objective To observe the effects of Tao-He-Cheng-Qi decoction on rat vascular expression of toll-like receptor-2 (TLR-2),TLR-4,transforming growth factor-beta (TGF β),and insulin-like growth factor-1 (IGF-1) expression.Methods Male SD rats were divided to a control group (A group,n=30),a model group (B group,n=31),an early-treatment group (C group,n=31) and a regular-treatment group (D group,n=31).Rats in A group had no drug intervention,but received 0.9% sodium chloride (10 mL/kg) gavage everyday in rats of B group.Rats in C group were received Tao-He-Cheng-Qi decoction by gavage before and after diabetic modeling.The rats of D group were received Tao-He-Cheng-Qi decoction after diabetes modeling.After 1,12,or 20 weeks of intervention,rats were sacrificed.Immunohistochemical methods were used to detect the expression of TLR-2,TLR-4,TGFβ and IGF-1 in large blood vessel.Results Twenty weeks after intervention,the levels of TLR-2 expression were (9.333± 2.338) pg/mL in D group,but significantly decreased in C group (3.750 ± 1.282 pg/mL,P < 0.05).The levels of TLR-2 expression were not different between D and B group.The levels of TL-4 expression were (3.750 ± 1.282) pg/mL in C group and (5.750± 1.982) pg/mL in D group,which was significantly decreased compared to that in B group (10.333 ± 2.658 pg/mL,P < 0.05).The expression levels of TGF-beta was (403.438 ± 11.273) pmol/L in C group and (535.313±12.384) pmol/L in D group,with significant difference (P< 0.05).The expression levels of IGF-1 were (1104.375±55.495) pmol/L in C group and (861.000±28.656) pmol/L in D group.The difference was significant (P < 0.05).Conclusion Tao-He-Cheng-Qi decoction can reduce diabetic vascular fibrosis,and also can prevent diabetic vascular fibrosis when early intervention.
