胰腺导管腺癌患者血清中microRNA-155的表达及其临床意义

摘要

目的 检测microRNA-155(miR-155)在胰腺导管腺癌(pancreatic ductal adenocarcinoma,PDAC)组织中、患者血清中的含量,探讨其作为PDAC血清学诊断标志物的可能性.方法 应用荧光实时定量PCR法检测miR-155在20对PDAC组织和癌旁组织、70例PDAC患者和40例健康人血清、10对荷人PDAC小鼠和对照组小鼠血清及6株PDAC细胞株和其培养上清中的含量.结果 实验结果 显示,PDAC组织中miR-155表达高于癌旁胰腺组织(P=0.005);并且miR-155在PDAC患者血清中表达量高于健康对照组(P=0.004);在荷人PDAC小鼠血清中miR-155含量高于对照组小鼠(P=0.012);同时,6株PDAC细胞株miR-155的表达存在差异(P=0.000),miR-155在细胞内和在其培养上清中的表达具有高度一致性(r=0.628,P=0.005).结论 miR-155在PDAC组织和患者血清中表达上调,提示其可能是PDAC血清学的潜在标志物.%Purpose To investigate the expression of mieroRNA-155 ( miR-155 ) in panereatic ductal adenocarcinoma ( PDAC ) tissue and serum, as well as diagnostic value of miR-155 in serum of PDAC. Methods Tissue specimens from PDAC and pancreas ( n =20 and n = 20 , respectively ), the sera of patients with PDAC and control group ( n =70 and n = 40 , respectively ), the sera of SCID mice with humans' PDAC and control group ( n = 6 and n = 6 , respectively ), the 6 PDAC cells and supernatant were analyzed via real-time RT-PCR for miR-155.Results The expression of miR-155 in PDAC tissue was significantly higher than that in pancreatic tissue ( P =0.005 ). The expression of miR-155 in the sera of patients with PDAC was significantly higher than that in control group ( P = 0.004 ).The expression of miR-155 in the sera of mice with humans' PDAC was significantly higher than that in control group ( P =0.012 ). It was been found different expression level of miR-155 among 6 pancreatic cell lines ( P = 0.000 ), a positive correlation was observed between the expression of miR-155 in the 6 PDAC cells and supernatant ( r = 0.628, P = 0.005 ). Conclusion The expression of miR-155 in the tissue and sera of patients with PDAC is significantly higher than that in control group. Therefore, serum miR-155 may be used as a potential biomarker in the diagnoses of PDAC.
