8-姜酚与麦芽糖基-β-环糊精的包合研究

         

摘要

8-gingerol is a biologically active component of ginger which contributes to human health.However,the applications of 8-gingerol is limited due to the low water solubility and poor stability.Cyclodextrin can be used as wall material of microcapsule for bioactive compounds to improve the water solubility and stability.The inclusion complexes of 8 gingerol with 6-O-α-maltosyl-β-cyclodextrin (Mal-β-CD) were prepared via solvent method and freeze-drying in this study.The inclusion behavior and interaction of 8-gingerol and Malβ-CD were characterized by UV,FT IR,XRD,TG/DSC and molecular docking simulations.All obtained information demonstrated the formation of the host-guest complex and suggest that 8-gingerol was encapsulated with the hydrophobic cavity of Mal-β-CD.The results also showed that the stoichiometry of the 8-gingerol/Malβ CD inclusion complex was 1:1,the binding energy was-4.4 Kcal/mol and hydrogen bond lengths was 2.9/(A).8-gingerol was completely dispersed in Mal-β-CD and its heat stability had been significantly enhanced.%8-姜酚是生姜中有益于人体健康的生物活性成分,但其水溶性较低,化学稳定性较差,这限制了其在食品工业中的应用.环糊精可作为生物活性成分微胶囊的壁材改善其水溶性和稳定性,本研究采用溶剂法联合冷冻干燥技术制备了8-姜酚/麦芽糖基-β环糊精(Mal-β-CD)的包合物,并借助紫外光谱、红外光谱、X射线衍射技术、热重及差示扫描量热联用分析、分子对接模拟等技术研究包合物结构特征.结果表明,Mal-β-CD对8-姜酚具有良好的包合能力,8-姜酚与Mal-β-CD包合物的化学计量比为1:1;8-姜酚的苯环结构进入到Mal-β-CD的疏水性空腔中,二者之间形成了氢键,结合能为-4.4 Kcal/mol,氢键键长为2.9 A.8姜酚以无定形状态存在于包合物中,其热稳定性得到了明显提高.
