海洋细菌0417产胞外多糖发酵条件的优化

     

摘要

微生物多糖依据其不同的特性被广泛的应用于多个领域.近年来,微生物多糖抗肿瘤、抗炎、抗病毒等生物活性愈来愈受到关注.海洋微生物因其独特的生活环境而使其胞外多糖具有独特的结构和功能.对从威海近海分离筛选到的具有产胞外多糖能力的海洋细菌0417进行发酵条件的优化探究.结果表明,海洋细菌0417产胞外多糖的最佳碳源、氮源分别为葡萄糖和蛋白胨;培养基初始pH值为7.5;碳氮源最佳质量浓度分别为10g/100mL和0.6g/100mL;胞外多糖积累最佳时间为120h.在此条件下培养,海洋细菌0417胞外多糖产量可达1.62mg/mL.%Microbial polysaccharide, according to their different characteristics, are widely used in many rields. In recent years, the biological activities of microbial polysaccharide, like anti-tumor, anti-inflammatory, anti-virus and so on attract more and more attention. Marine microorganisms feature a unique exocellular polysaccharide structure and function owing to its unique environment. The marine bacterium 0417, which has a production capacity of extracellular polysaccharide, was isolated from the seawater samples from Weihai offshore and was chosen for exploring the optimization of fermentation conditions. Results showed that the better carbon and nitrogen sources for marine bacterium 0417 to produce extracellular polysaccharide were glucose and peptone, respectively. The initial pH value of the medium was about 7.5; the optimal mass concentrations of carbon and nitrogen sources were 10g/100ml and 0.6g/100ml, respectively. The optimal harvest time for extracellular polysaccharide was about 120h. Under such conditions the marine bacterium 0417 could produce 1.62mg/ml exopolysaccharide.
