气相色谱法测定丙酮丁醇梭菌发酵液中的代谢产物及糠醛

     

摘要

A quantitative analysis method was established to determine the products of the acetone-butanol fermentation including acetone, ethanol, butanol, butyric acid and furfural by gas chromatography, in which isobutanol was used as an internal standard and FFAP (30m×0.25mm×0.33μm) was used as the analytical column and FID as the detector. The sample was separated by two-step programmed temperature method. The carrier gas was nitrogen, and the pre-column pressure was 0.12MPa. The split ratio was 40:1 and the injection volume was 0.2μL. The experimental results showed that components could be completely separated and had a good linear relationship between the concentration and peak area ratio. The relative standard deviations were no more than 1.65%, and the average recoveries were from 91.42% to 108.01%. This method was available for the determination of product in the acetone-butanol fermentation. .%该文建立了一种测定丁醇发酵液中丙酮、乙醇、正丁醇、丁酸等主要代谢产物及糠醛含量的气相色谱分析方法.以异丁醇为内标,色谱条件为:FFAP毛细管柱(30mx0.25mmx0.33μrn),FID检测器;柱温:初始温度60°C,保持1min,以1OC/min升至110°C,保持4min,再以15°C/min升至150℃,保持3min.载气:氮气,柱前压为0.12MPa;分流比:40:1;进样量:0.2μL.结果表明,各组分能完全分离,各组分浓度与峰面积比线性关系良好,线性相关系数≥0.9980,重复性相对标准偏差RSD≤1.65%,各组分3种不同浓度加标回收率在91.42%~108.01%之间.该法具有前处理容易、进样量少、分析组分多、线性范围宽、简便快捷等优点,可同时对丙酮丁醇梭菌发酵过程中的多种组分进行有效分析.
