The extraction process of nucleoprotein and protamine from freshwater fish spermary was studies. The extraction conditions were optimized by using the orthogonal design. In extracting nucleoprotein, NaCl concentration was 1 .0mol/L, the volume of NaCl solution was ten times as large as that of fish spermary, extraction time was 1 hour, the volume of 95% ethanol solution was two times as large as that of fish spermary. In extracting protamine, H_2SO_4 concentration was 5.0%, the volume of H_2SO_4 and 95 % ethanol solution were three times as large as those of fish spermary, extraction time was two hours. Simultaneously, the nutritive values of nucleoprotein and protamine were analysed. The results showed that the characteristics of nucleoprotein compositions were higher in DNA, arginine and zinc, and reasonable essential amino acids pattern. Therefore, nucleoprotein was a good material for curing sterility for man and resisting aging. The protein content of protamine was 92.8%, and the contents of arginine was higher than that in nucleoprotein. So the protamine is a kind of fine protein and drug sources for curing man's sterility. Moreover, it is an effective natural antiseptic as well.
