Methods and compositions are provided involving universal sequencing primers. The primer is characterized by having a double-stranded region, joined by a uniquely cleavable linkage and capable of being linked to a single strand of DNA. One or two universal linkers may be employed for joining to one or both ends of the ds DNA, whereupon denaturation, two extended single-stranded sequences may be obtained terminating in the universal sequencing primer linker ("splinker"). Each of the strands may then be sequenced by conventional methods employing template recognition.
