method for modifying a targeted site in a double-stranded DNA of a gram-positive bacterium, nucleic acid-modifying enzyme complex, and nucleic acid
the present invention provides a method for modifying a targeted double-stranded DNA site owned by a gram-positive bacterium, the method comprising a step of placing a complex, in which a nucleic acid-based converting enzyme is attached to a nucleic acid sequence recognition that specifically binds to a target nucleotide sequence in selected double-stranded DNA, in contact with the double-stranded DNA, to delete or convert at least one nucleotide in the targeted site into another at least one nucleotide, or insert at least one nucleotide in the targeted site, without cleavage of at least one double-stranded DNA strand in the targeted site, where the contact of the double-stranded DNA with the complex is accomplished by introducing a nucleic acid, which encodes the complex, in gram-positive bacteria. the present invention also provides a complex that is used in the method and wherein a nucleic acid-based converting enzyme is linked to a nucleic acid sequence recognition module that specifically binds to a target nucleotide sequence in double-stranded dna possessed by gram-positive bacteria.
