The present invention relates to a pharmaceutical composition for preventing or treating brain diseases, comprising a KDM4B inhibitor as an effective component. In particular, the present invention has confirmed that when inflammation reactions were induced by TNF in human brain microvascular endothelial cells, the methylation of ICAM1 promotor regions (H3K9me2 and H3K27me3) was reduced; the inhibition of histone methyltransferase G9a increases ICAM1 expression by reducing H3K9me2; and the overexpression of G9a suppresses ICAM1 expression, thus confirming that the methylation of ICAM1 promotor region regulates ICAM1 expression. Further, lysine(K)-Specific Demethylase 4B (KDM4B), which is a histone demethylase, binds to the ICAM1 promotor regions; and the inhibition of KDM4B expression suppresses ICAM1 expression, suppresses white blood cell adhesion, and increases the expression of A protease, neprilysin, and thus the KDM4B inhibitor of the present invention may be beneficially used as a pharmaceutical composition for preventing or treating brain diseases.
