Studies on Improving and Standardizing the Process for Preparation of Cryoprecipitated Antihemophilic Factor (Human).

Storage of ACD blood at 4 C for 6and 24hours resulted in a decrease of the Factor VIII content of both the cryoprecipitate and the parent plasma relative to activity of the freshly drawn preparation. Calculation of Factor VIII recovery (in vitro) showed a significant reduction in Factor VIII activity when the storage time of the blood was prolonged from less than 1hour to 6hours,but not when the storage time was prolonged from 6hours to 24hours. The project director also found that the cryoprecipitate and the parent plasma seem to have higher Factor VIII activity when activity is determined by the thromboplastin generation test than when it is determined by the partial thromboplastin time.
