Acute Cardiovascular Effects of Coralyne Sulfoacetate (NSC-154890) Following Single Bolus Intravenous Injections to Anesthetized Beaglehounds Denied Heparin.

Effects of 32mg Coralyne/kg body weight on cardiovascular parameters of six anesthetized, unheparinized dogs were studied by analysis of ECG's,pressures from the aorta,pulmonary trunk and left ventricle,cardiac output,and blood gases during a 60-minute period following intravenous injection. Heart rate and respiratory rates both increased markedly,left ventricular contractility,aortic and pulmonary trunk pressures,and cardiac output fell drastically during the immediate post-injection period. Contractility,alone,returned towards normal by 60minutes post-infusion PaO2and PaCO2fell slightly,but pH did not change. QT interval shortened in concert with tachycardia,and ST-T alterations indicating possible left ventricular endocardial ischemia developed along with large voltage T-waves in all leads. By 90minutes post-injection,one dog died of shock (hypotension and low cardiac output) while a second dog died with massive intravascular clots.
