Competitive Inhibitor-Enzyme Immunoassay

Components for a competitive inhibitor-enzyme immunoassay were prepared and tested. Hapten-inhibitor conjugates were prepared and evaluated using both penicillin and phenobarbitol as haptens. Penicilloyl-phenylbutylamine, when bound to an antibody, was not bound by chymotrypsin to a significant extent so was not useful. Phenobarbital-valeryl-D-tryptophan methyl ester, when bound to antibody specific for phenobarbital was bound by chymotrypsin to a slight, but not useful, extent. Phenobarbital-octanoyl-D-tryptophan methyl ester, when bound to anti-phenobarbital antibody, was bound by chymotrypsin to a potentially useful extent. Photoreactive derivatives of chymotrypsin were also successfully prepared after problems of instability and aggregation were solved. It is concluded that competitive inhibitor-enzyme immunoassay will be shown to be feasible.
