Tributyrin agar, selective media for isolation of lipase producing bacteria (Bacillus subtilis) was used. Lipase producing microorganisms produced clear zones on this media. Agar disc cut from the clear zone also show the clear zone when placed on thefresh selective medium to get the pure culture. A comparative study on the production of extra cellular lipase by Solid State Fermentation (SSF) using Bacillus subtilis with various substrates has been made. Different parameters such as substrate selection, pH of the medium, incubation time, effect of moisture content of substrate were optimized. The maximum extracellular lipase activity of 4.65 units per gram of dry fermented substrates (U g ds~(-1)), 4.62 U g ds~(-1) with initial moisture content of70% and 4.71Ug ds~(-1)at pH8 was observed with Coconut oil cake as a substrate.
