To search for additional genomic components which may be necessary for ageratum yellow vein bigeminivirus (AYVV) infection of A. conyzoides, naturally-infected A. conyzoides plants were collected from wastelands in Singapore. Virus-specific supercoiled DNA was purified by CsCl centrifugation and clones containing viral DNA inserts were identified by colony hybridization. The supercoiled DNA did not contain detectable levels of a DNA B component, but novel defective DNAs associated with infected A.conyzoides were identified. The defective DNA had a significant effect on AYVV DNA accumulation and symptom development when reintroduced into Nicotiana benthamiana by inoculation.
