InordertoknowtheroleoftheemXdsg/emgeneinpresumptivePGCs(pPGCs)ofemXenopus/em,weattemptedtoinhibitthetranslationofemXdsg/emmRNAinpPGCsbyinjectingantisensemorpholinooligo(asMO),togetherwithFluoresceinDextran-Lysine(FDL),intosinglegermplasm-bearingcellsof32-cellembryos.AmongthreetypesofasMOscomplementarytodifferentpartsofthe5prime;-untranslatedregionofemXdsg/emmRNAtested,onlyoneasMO,designatedasXdsg-3,inhibitedthetranslationofthemRNAinFDL-labeledpPGCs,resultingintheabsenceoflabeledPGCsinexperimentaltadpoles.Ontheotherhand,twootherasMOs,Xdsg-1and-2,didnotinhibitthetranslation,sothatasimilarnumberoflabeledPGCsfoundinFDL-injectedbutasMO-uninjectedcontroltadpoleswereobservedinexperimentaltadpolesderivedfromasMO-injectedembryos.Surprisingly,useofXdsg-3asMOresultedinthedisappearanceoftheproteinofemXenopusvasa/emhomolog(emXenopusvasa-likegene1/em,emXVLG1/em)fromFDL-labeledpPGCsbyinhibitingthetranslationofemXVLG1/emmRNA.However,theeffectofXdsg-3asMOonthetranslationofemXdsg/emandemXVLG1/emmRNAsandPGCformationcouldbecanceledbythecoinjectionwithemXdsg/emmRNA.Consequently,theXdsgproteininpPGCsmayplayanimportantroleintheformationofPGCsbyregulatingtheproductionofXVLG1protein./p/div
