广西甘薯品种的茎尖诱导培养研究初报

             

摘要

Suitable culture conditions in vitro by stem tip inducing for commonly-grown sweet potato varieties in Guangxi were studied in this research. Popular cultivars Guifen No. 2 (high starch content) and Guishu 131 (edible purple sweet potato) were used as test materials. Then, using the stem tip culture method in vitro based on the MS culture medium, the disinfection and composition times under different 6-BA and NAA concentrations and the shoot tip survival rate, differentiation rate, plantlet survival rate, and seedling transplanting survival rate were studied. Results showed that the optimal duration for Guifen No. 2 disinfection was 5 minutes, and the stem tip survival rate was 76. 0%. On the other hand, the optimal duration for Guishu 131 sterilization was 4 minutes, and the stem tip survival rate was 74.4%. For Guifen No. 2, the best culture medium was solid MS+6-BA 2. 0mg/L+NAA 0.lmg/L, the stem tip differentiation rate was 85.29% , the plantlet survival rate was 64.71% , and the transplanted plantlet survival rate was 90. 91%. Meanwhile, the suitable culture medium for Guishu 131 was solid MS+6-BA 1.5mg/L+NAA 0.1 mg/L, the stem tip differentiation rate was 80.95% , plantlet survival rate was 80. 95% , and the transplanted plantlet survival rate was 100.00%. The corresponding culture conditions selected for Guifen No. 2 and Guishu 131 could be applied for the production of virus-free sweet potato seedlings.%研究适合广西主栽甘薯品种的茎尖诱导培养条件.以广西主栽甘薯品种长蔓基因型桂粉2号(高淀粉)和短蔓基因型桂薯131(食用紫薯)为材料,用茎尖离体诱导培养方法在MS基本培养基的基础上研究消毒时间及复合添加不同浓度6-BA和NAA对茎尖存活率、分化率、成苗率及移栽成活率的影响.结果表明,桂粉2号的最佳消毒时间为5min,茎尖存活率为76.0%;桂薯131的最佳消毒时间为4min,茎尖存活率为74.8%.桂粉2号的最佳培养基为固体MS+ 6-BA 2.0 mg/L+ NAA0.1 mg/L,其茎尖分化率为85.29%,成苗率为64.71%,移栽成活率为90.91%;桂薯131的适宜培养基为固体MS+ 6-BA 1.5 mg/L+ NAA0.1mg/L,其茎尖分化率为80.95%,成苗率为80.95%,移栽成活率100.00%.筛选出的培养基可用于桂粉2号及桂薯131规模化甘薯无毒苗生产.
