目的 探讨热休克处理在肿瘤坏死因子-α(TNF-α)诱导胃腺癌BGC823细胞凋亡过程中的作用,为胃癌的生物学治疗提供依据.方法 将体外培养的胃腺癌BGC823指数生长期细胞分为热休克组和对照组,热休克组行热休克处理.两组均分别用0、25、50、100 ng/m1浓度的TNF-α诱导,作用4 h;热休克组行热休克处理.采用TUNEL法和3H-TdR掺入试验检测两组肿瘤细胞凋亡指数(AI)及细胞增殖抑制率(IR).结果 与对照组比较,热休克组AI明显下降(P<0.01);IR明显升高(P<0.01),呈剂量依赖性.结论 热休克处理对TNF-α诱导的胃腺癌BGC823细胞凋亡具有抑制作用;此作用与TNF-α呈剂量依赖效应.%Objective To investigate the effect of heat shock treatment the apoptosis of gastric adenocarcinoma cells BGC823 induced by TNF-α. Methods Gastric adenocarcinoma cells cultured in vitro were divided to heat shock group and control group ,an then treated by TNF-α of 0,25,50,100 ng/ml for 4 hs. TUNEL and 3H-TdR test were used to examine apoptosis index and DNA synthsis. Results Compared with the control group,the apoptosis index of heat shock group decreased significantly( P <0.01 ) ;and the cpm significantly increased( P <0. 01 ) ,which showed dose-depended manner.Conclusions Heat shock treatment can inhibit the apoptosis of gastric adenocarcinoma cells induced by TNF-α in vitro;this effect showed dose-depended manner.
