不同食管病变组织中TGFβ1、α-SMA、smad4、VEGF蛋白表达及MVD比较

         

摘要

Objective To detect and compare the changes of transform growth factor β1 (TGFβ1),α-SMA, smad4 protein, vascular endothelial growth factor ( VEGF) and microvessel density ( MVD) in the normal esophageal tissues, low-grade intraepithelial neoplasia ( LGIEN) , high-grade intraepithelial neoplasia ( HGIEN) , early esophageal cancer and ad-vanced esophageal cancer tissues.Methods Twenty cases of normal esophageal tissue specimens ( NC group) , 30 cases of LGIEN specimens ( LGIEN group) , 50 cases of HGIEN specimens, 25 cases of early esophageal cancer specimens ( CIS group) and 25 cases of advanced esophageal cancer specimens (SCC group) were selected.The expression of TGFβ1,α-SMA, smad4 and VEGF protein in each group was detected by immunohistochemistry, and the MVD was determined by Antibody CD34.Results From NC group to SCC group, the positive expression rates of α-SMA, TGFβ1 and VEGF and MVD were gradually increased (all P<0.05);the positive expression rate of smad4 protein was gradually decreased (all P<0.05);and the positive expression rate of TGFβ1 was first increased and then decreased, and the CIS group was the highest (all P<0.05).Conclusions In the course of the development of esophageal cancer from normal esophagus to ad-vanced stage, the phenotype and function of esophageal stromal fibroblasts change, and TGF β1/smad4 protein pathway and angiogenesis may play an important role in the protein pathway.%目的 比较正常食管组织、低级别食管上皮内瘤变、高级别食管上皮内瘤变、早期食管癌和进展期食管癌组织中TGFβ1、α-SMA、smad4、VEGF蛋白表达及微血管密度(MVD).方法 选取正常食管组织标本20份(NC组),低级别食管上皮内瘤变组织标本30份(LGIEN组),高级别食管上皮内瘤变组织标本50份(HGIEN组),早期食管癌组织标本25份(CIS组)和进展期食管癌组织标本25份(SCC组),采用免疫组化法检测各组标本中的TGFβ1、α-SMA、smad4、VEGF蛋白,用CD34标记血管内皮细胞测算各组MVD.结果 从NC组到SCC组,食管间质成纤维细胞α-SMA、TGFβ1、VEGF阳性表达率和MVD逐渐升高(P均<0.05);上皮细胞smad4蛋白阳性表达率逐渐下降(P均<0.05),上皮细胞TGFβ1阳性表达率先升后降,CIS组最高(P均<0.05).结论 随着正常食管组织向进展期食管癌组织转变,其中间质成纤维细胞的表型和功能发生改变,TGFβ1/smad4蛋白通路和血管新生可能在其中发挥重要作用.
