均匀设计优化葡萄糖氧化酶发酵培养基

     

摘要

为了提高黑曲霉发酵生产葡萄糖氧化酶的产率,采用单因素实验考察碳源和氮源的种类对产酶的影响,然后采用均匀设计对发酵培养基的碳源、氮源、KH2PO4、MgSO4·7H:0、CaCO,的浓度水平进行考察,实验数据进行二次多项式逐步回归模型分析。结果显示,葡萄糖、(NH。):HPO。对产酶影响效果显著;发酵培养基的最佳组成为:葡萄糖16.24%、(NH4)2HP040.12%、KH2PO。0.57%、MgS04。7H20O.63%、CaC035.69%。%In order to improve the production of glucose oxidase by Aspergillus niger, the effect of carbon source and nitrogen source of fermentation medium were explored in single factor experiments and the concentration level of carbon source, nitrogen source, KHzPO4, MgSO4 · 7H20 and CaCO3 were studied though uniform design methodology.The results indicated that. glucose and (NH4 )2HPO4 of enzyme effect was remarkable.The optimum fermentation medium for production of glucose oxidase was composed of glucose of 16.24%, ( NH, )2HPO4 of 0. 1 2 %, KH2 PO4 of 0.57 %, MgSO4 · 7 H20 of 0.63 %, CaCO3 of 5.69 %.
