温石棉及其代用纤维对鼠细胞钙激活蛋白43及B 淋巴细胞瘤-2表达的影响

         

摘要

Objective To investigate the effect of chrysotile and its substitute on expressions of CAP 43 and Bcl-2 in hamster cells and to explore the their possible carcinogenic mechanism .Methods Chinese hamster lung cells(V79 cells)were exposed to dif-ferent concentrations of chrysotile and its substitutes including chrysotile from Xinkang ,Sichuan(A),chrysotile from Southern Shanxi (B),glass fiber(C),ceramic fiber(D),wollastonite(E)and rockwool(F).At the same time,cells without treatment were used as nega-tive controls.The expressions of CAP43 and Bcl-2 in V79 cells were detected by using immunohistochemistry ,and compared by using statistical program SPSS21.0.Results The expressions of CAP43 and Bcl-2 in A and B were significantly up-regulated(P <0.05). As the concentration of powder increased ,immune staining OD value of CAP 43 and Bcl-2 were increased .Conclusion Chrysotile and its substitute can significantly influence the CAP 43 and Bcl-2 expressions in hamster cells .%目的:研究温石棉及其代用纤维对鼠细胞钙激活蛋白43(calciumactinated protein 43,CAP43)及B淋巴细胞瘤-2(B-cell lymghoma,Bcl-2)表达的影响,探讨温石棉及其代用品的致癌性机制。方法将不同浓度的温石棉及其代用品[四川新康温石棉(A)、陕西陕南温石棉(B)、玻璃纤维(C)、陶瓷纤维(D)、硅灰石(E)、岩棉(F),阴性对照组(G)]暴露于V79细胞,使用免疫组织化学法检测CAP43及Bcl-2的表达,并使用SPSS 21.0对表达结果进行对比分析。结果 A、B组的仓鼠肺细胞中CAP43及Bcl-2表达增强较为显著,与其他组比较,差异有统计学意义( P<0.05);CAP 43及Bcl-2基因表达随着粉体浓度的增加,其OD值也呈上升趋势。结论温石棉及其代用纤维对于鼠细胞CAP43及Bcl-2表达有显著的增强作用。
