乳腺癌中VEGF基因mRNA的表达及其与临床病理因素的关系

             

摘要

目的 探讨乳腺癌组织中血管内皮生长因子(vascular endothelial growth factor,VEGF)mRNA表达情况及其与临床病理因素及其他分子指标的关系.方法 应用实时荧光定量逆转录-聚合酶链反应检测48例乳腺癌组织,20例乳腺纤维腺瘤组织VEGF mRNA的表达,并以免疫组化SABC法检测VEGF蛋白在乳腺癌、乳腺纤维腺瘤组织中的表达,分析其与乳腺癌中HER-2,ER,PR蛋白表达的关系.结果 经实时荧光定量RT-PCR法检测,VEGF121mRNA、VEGF165mRNA在乳腺癌组织,乳腺纤维腺瘤组织中的2-ΔCt值分别为:0.5733±0.1764、0.310±0.098,0.2961±0.0845、0.2657±0.0342.乳腺癌组织的VEGF mRNA阳性表达率均显著高于乳腺纤维腺瘤组织(P<0.05),但与患者的年龄、淋巴结转移、肿块大小、组织学分级、pTNM分期等临床病理因素无关(P>0.05).HER-2在乳腺癌和纤维腺瘤中表达分别为18.8%、0%,差异具有显著性(P<0.05).Spearman等级相关分析表明VEGF与HER-2,ER,PR表达不相关(P>0.05).HER-2与雌激素受体、孕激素受体的表达呈负相关(γ=-0.335,P=0.005;γ=-0.300,P=0.012).结论 VEGF在乳腺癌中高表达,但与ER,PR表达不相关.HER-2可作为判断乳腺癌预后的1个重要参考指标.%Objective To investigate the expression of VEGF in breast cancer tissue and its relation to clinicopathological and other molecular parameters. Methods The expression of VEGF mRNA in 48 cases of breast carcinoma and 20 cases of breast fibroadenoma issues were detected by fqRT-PCR and the protein expression of VEGF in those issues were detected hy SABC immunohistochemistry. The protein expression of HER-2, ER. PR in 48 cases of breast carcinoma were detected by using SABC immunohistochemistry. The relationship of its expression with VEGF,HER-2 ,ER, PR were analyzed. Results By RT-PCR method , the 2-ΔCt values of VEGF121 mRNA and VEGF165 mRNA of the breast carcinoma and breast fibroadenoma tissues were 0. 5733 ±0. 1764,0. 310 ±0. 098 and 0. 2961 ±0. 084,0. 2657 ±0. 0342 respectively. The positive expression rates of VEGF mRNA were significantly higher in breast carcinoma than those in breast fibroadenoma tissues,there were significant difference among the two groups( P <0. 05 ). The positive expression rates of VEGF in hreast carcinoma had no significant correlation with the age,lymph node metastasis , tumor size , differentiation degree , pTNM stages of the patient( P > 0. 05 ). The expression of HER-2 were 18. 8% and 0% in breast carcinoma and fibroadenoma issues respectively. there were significant difference among the two groups( P < 0. 05 ). The expression of HER-2 were negatively associated with that of ER and PR( γ =- 0. 335 ,P = 0. 005 ; γ =-0. 300 ,P =0. 012 ). The expression of VEGF had no correlation with that of HER-2 ,ER,PR ( P > 0. 05 ) by Spearman analysis. Conclusion Overexpression VEGF mRNA is over - expressed and not correlated with the ER,PR status. HER - 2 may serve as an important prognostic factor of breast cancer
