Objective To observe the inhibitory effect of OX26 and As2 O3 immunoliposome on viability of glioma cell in rats in vitro.Methods C6 glioma cells were cultured in vitro and divided into saline water group,As2 O3 group,and combination group of OX26 and As2 O3 immunoliposome.MTT colorimetric method was used to detect inhibitory effect of combination of OX26 and As2 O3 immunoliposome on viability of glioma cells.DAPI staining was used to observe apoptosis of combination of OX26 and As2 O3 immunoliposome on glioma cells.Western blot method was used to evaluate expression of glial fibrillary acidic protein ( GFAP) combination of OX26 and As2 O3 immunoliposome on glioma cells.Results MTT method showed inhibitory rate of com-bination of OX26 and As2 O3 immunoliposome on glioma cells was 40.21%,which was higher than As2 O3 group 25.77%( P<0.05).Compared with saline water group and As2O3 group,optical density and areal density of glioma cells in combination group of OX26 and As2O3 immunoliposome were obviously decreased by DAPI staining(P<0.01).Western blot method showed that GFAP expression in combination group of OX26 and As2 O3 immunoliposome was less than saline water group and As2 O3 group. Conclusion Inhibitory effect of combination of OX26 and As2 O3 immunoliposome on viability of glioma cell rats in vitro is re-markable,and its mechanism may be related with promote apoptosis and GFAP expression of glioma cells.%目的：探讨转铁蛋白受体单克隆抗体（ OX26）偶联三氧化二砷（ As2 O3）免疫脂质体对鼠脑胶质瘤细胞株的体外抑制作用。方法体外培养C6脑胶质瘤细胞，实验分为盐水组，As2 O3（6μmol· L－1）组和OX26偶联As2 O3脂质体（6μmol· L－1）组；采用四唑盐比色法（MTT），检测OX26偶联As2O3免疫脂质体对脑胶质瘤细胞生长的抑制作用， DAPI染色观察OX26偶联As2 O3免疫脂质体对脑胶质瘤细胞促凋亡的影响。免疫印迹法检测OX26偶联As2 O3免疫脂质体对脑胶质瘤细胞胶质瘤胶质纤维酸性蛋白（ GFAP ）蛋白表达的影响。结果 MTT比色法检测显示：OX26偶联As2 O3免疫脂质干预对脑胶质瘤生长的抑制率为40．21％，显著高于As2 O3组的25．77％，比较差异有统计学意义（ P＜0．05）；核染显示：与盐水组和As2 O3组比较，OX26偶联As2 O3免疫脂质组脑胶质瘤细胞的光密度和面密度值显著减少，比较差异有统计学意义（P＜0．01）；Westernblot检测显示：OX26偶联As2O3免疫脂质组脑胶质瘤细胞GFAP蛋白的表达较盐水组和As2 O3组少。结论 OX26偶联As2 O3免疫脂质体对鼠脑胶质瘤细胞具有明显的体外抑制作用，其作用机制可能与促进脑胶质瘤细胞凋亡和GFAP蛋白的表达相关。
