目的:建立威麦宁胶囊中原花青素B2、表儿茶素的HPLC测定方法.方法:采用高效液相色谱法测定原花青素B2、表儿茶素的含量,其标准曲线、精密度、重复性、加样回收率均符合要求.色谱柱:Zorbax SB-C18柱,流动相为乙腈-0.04％磷酸水溶液,梯度洗脱,检测波长为280nm,柱温30℃,流速1.0 mL·min-1,进样量10 μL.结果:原花青素B2、表儿茶素的标准曲线分别为A原花青素B2=7047.3C-1066 (r=0.9998),A表儿茶素=7321.9C-601.43 (r=0.9999),加样回收率分别为93.56％(RSD 4.36％)、99.34％(R SD 4.14％),3个批次威麦宁胶囊中原花青素B2、表儿茶素的平均含量分别为1.44mg/粒、0.96mg/粒.结论:威麦宁胶囊3个批次中2种主要成分的含量相近.该方法简便、准确、重复性好,可为威麦宁胶囊提供质量控制方法.%Objective: To establish an HPLC method for the determination of procyanidin B2 and epicatechin in Weimaining capsules. Methods: An HPLC method was established with its standard curves, precisions, repeatabilities and recoveries met to the requirements. The determination was performed on a Zorbax SB—C18 column. The mobile phase was consisted of acetonitrile —0.04% phosphoric acid (gradient elution). The detection wavelength was set at 280 nm and the column temperature was 30℃. The flow rate was 1.0 mL·min-1. The injection volumn was 10 μL. Results: The standard curves were Aprocyaindin B2 = 7047.3C-1066 (r=0.9998) and Aepicatechin=7321.9C-601.43 (r=0.9999) with average recoveries at 93.56% (RSD 4.36%) and 99.34% (RSD 4.14%), respectively. The average content of procyanidin B2 and epicatechin in 3 batches of Weimaining Capsules were 1.44 mg and 0.96 mg per capsule, respectively. Conclusion: The contents of procyanidin B2 and epicatechin in 3 batches of Weimaining Capsule are similar. The method is simple, accurate and reproducible for quality control of Weimaining capsules.
