芦丁对β淀粉样肽纤维化及细胞毒性的抑制作用

         

摘要

Objective To investigate the inhibitory effect of rutin on amyloid fibrillation and fibrillar cytotoxicity of Aβ25‐35 fragment . Methods Aβ25‐35 was incubated at pH 7 .4 and 37 ℃ to prepare amyloid fibrils .Fibril formation was monitored by thioflavin T (ThT) fluorescence and the fibrillar morphology was visualized under transmission electron microscopy .The inhibitory role of ru‐tin on amyloid fibrillar cytotoxicity was evaluated by measuring the cell viability of PC 12 cells with fibrils .Results Incubation of Aβ25‐35 fragment at pH 7 .4 and 37 ℃for 60 h resulted in the formation of amyloid fibrils .Rutin inhibited amyloid formation ,disrup‐ted the fibrillar structure and decreased the cell damages induced by Aβfibrils .Conclusion Rutin is effective to inhibit amyloid for‐mation of Aβ25‐35 fragment ,disrupt the structure of mature fibrils and attenuate the Aβfibril‐induced cytotoxicity .%目的:探索芦丁对Aβ25‐35肽段淀粉样纤维化及纤维细胞毒性的抑制作用。方法在pH值为7.4、温度37℃孵育Aβ25‐35肽,采用硫黄素(thioflavin T ,ThT)荧光和透射电子显微镜检测多肽的淀粉样纤维化;以淀粉样纤维处理PC12细胞建立的细胞损伤模型,M T T法检测细胞存活率,以评估芦丁对β淀粉样纤维细胞毒性的抑制作用。结果 Aβ25‐35肽段在p H值为7.4、温度37℃条件下,经孵育60 h左右形成淀粉样纤维;芦丁抑制Aβ淀粉样纤维的形成,破坏纤维结构,并降低纤维诱导的细胞损害。结论芦丁能够抑制Aβ25‐35的淀粉样纤维化和破坏成熟纤维结构,并降低Aβ纤维的细胞毒性。
