响应面法优化灵芝菌丝体胞内灵芝酸的提取

         

摘要

In this work, single factor experiment and response surface methodology were successfully applied to optimize the operational parameters which affected the extraction yields of ganoderic acids from the mycelia of Ganoderma lucidum. A quadratic surface model based on 3-levels, 3-factors central composite design (CCD) was employed to describe the relationship between IGA yields and operational parameters. The maximum extraction yields were 28.36 mg IGA per gram dry cell weight (DCW) ,when the mycelia were extracted with the following operational parameters:ethanol concentration 93.6% (v/v) ,temperature 79. 1 ℃ ,liquid-solid ratio 42.2:1 ,extraction time 2 h,extraction times 2. Confirmatory experiments under the optimum operational parameters showed IGA yields of 28.72 mg/g DCW which were in fine agreement with the theoretical yields of 28.36 mg/g DCW.%利用单因子试验和响应面法优化了影响灵芝菌丝体灵芝酸的提取过程.基于3因素3水平的中心组合设计,得到了描述胞内灵芝酸提取得率与操作参数之间的二次响应面模型.在乙醇浓度为93.6%(v/v)、提取温度79.1 ℃、提取液固比42.2∶1、提取次数2次、每次提取时间2 h的条件下,最大灵芝酸理论提取得率为28.36 mg胞内灵芝酸每克干菌丝.模型优化条件下的实际灵芝酸提取得率(28.72 mg/g干菌丝)与理论灵芝酸提取得率(28.36 mg/g于菌丝)相符.
