通络玉液汤对糖尿病大鼠大动脉血管内皮细胞超微结构和功能的影响

         

摘要

目的 探讨通络玉液汤对糖尿病大鼠大动脉血管内皮细胞超微结构和功能的影响.方法 选择72只SD大鼠进行实验,随机取24只大鼠作为正常组,给予普通饲料喂养;其余大鼠给予高糖高脂饲料喂养4周,第5周禁食12h后给予一次性腹腔注射链脲佐菌素造模,正常组给予等量柠檬酸钠缓冲液腹腔注射.将造模成功糖尿病血管病变模型大鼠随机分为模型组和通络玉液汤组,每组24只.通络玉液汤组给予通络玉液汤17 g/(kg·d)灌胃,正常组和模型组给予等量生理盐水灌胃,1次/d.在灌胃4周、8周时,每组各取12只,采用荧光法测定空腹动脉血一氧化氮(NO)和内皮素-1(ET-1)水平,透射电镜观察腹主动脉内皮细胞变化.结果 灌胃4周、8周后,与正常组比较,模型组和通络玉液汤组NO水平均明显降低(P均<0.05),ET-1水平均明显升高(P均<0.05);与模型组比较,通络玉液汤组NO水平均明显升高(P均<0.05),ET-1水平均明显降低(P均<0.05).灌胃8周后,模型组NO水平明显低于灌胃4周后(P<0.05),通络玉液汤组明显高于灌胃4周后(P<0.05);模型组ET-1水平明显高于灌胃4周后(P<0.05),通络玉液汤组明显低于灌胃4周后(P<0.05).电镜下,模型组内皮细胞表面突起、脱落,胞质内吞噬小泡,线粒体部分空泡化,核不规则,内皮细胞游离面胞质部分缺损,灌胃8周时比4周时更严重;通络玉液汤内皮细胞未见胞质脱落,胞质肿胀情况明显较轻,核规则,内质网完整.结论 通络玉液汤对糖尿病导致的大动脉血管内皮细胞结构和功能损伤有一定的保护作用.%Objective It is to explore the effects of Tongluoyuyetang (TLYY)on vascular endothelial ultrastructure and function of large artery in diabetic rats.Methods Seventy-two SD rats were selected,in which 24 ones as normal group fed with normal forage,the other ones were fed with high fat and sugar forage for 4 weeks,on the 5th week,they were given streptozocin by intraperitoneal injection to establish diabetic models,the ones in normal group were given the same dose of sodium citrate buffer solution by intraperitoneal injection.The successful models of diabetic angiopathies were randomly divided into model group and TLYY group,each group had 24 rats.TLYY group was given TLYY 17 g/(kg · d) by lavage,normal group and model group were given the same dose of normal saline by lavage,once per day.12 rats were selected respectively from each group after treatment for 4 and 8 weeks,and their serum NO and ET-1 was detected by fluorescence method,the changes of endothelial cells in man artery were observed by electron microscope.Results After treatment for 4 and 8 weeks,the levels of NO was decreased and the level of ET-1 was increased significantly in model group and TLYY group compared with that in normal group(P < 0.01);compared with model group,the level of NO was higher while that of ET-1 was lower in TLYY group (P < 0.05).After treatment for 8 weeks,the level of NO was lower in model group while the level in TLYY group was higher than that after treatment for 4 weeks (P < 0.05),the level of ET-1 was higher in model group while the level in TLYY group was lower than that after treatment for 4 weeks (P < 0.05).By electron microscopy,partial detachment of the vascular endothelial cells,partial deletion of nuclear membrane,vacuolation of the mitochondrion,destruction of the mitochondrial crista and more phagocytic vesicles were revealed in model group;and in TLYY group,vascular endothelial cell slight swelling,no loss of cytoplasm,nuclear regulation,endoplasmic reticulum integrity were found.Conclusion Tongluo Yuyetang have a certain protecting therapeutic effects on the damage of vascular endothelial structure and function caused by diabetes.
