空泡型质子泵与转化生长因子-β1在胃癌中表达及意义

         

摘要

目的:探讨空泡型质子泵(V-ATPases)与转化生长因子-β1(TGF-β1)在胃癌组织中的表达,并探讨其临床意义。方法:运用免疫组化方法检测胃癌组织及癌旁组织中V-ATPases与TGF-β1的表达水平,并分析两者相关性及与胃癌临床病理参数之间的关系。结果:免疫组化结果显示,(1)V-ATPases表达胃癌组织45例中阳性34例(75.5%),癌旁组织阳性13例(28.8%),两者比较差异有统计学意义(P<0.05)。(2)TGF-β1表达胃癌组织阳性35例(77.8%),癌旁组织阳性15例(33.3%),两者比较差异有统计学意义(P<0.05)。(3)相关性分析结果:V-ATPases的表达与TGF-β1的表达呈正相关(P<0.05);V-ATPases与TGF-β1的表达与病理分级、淋巴结转移、TNM分期、脉管浸润呈正相关(P<0.05),与临床病理特征如性别、年龄、肿瘤大小无相关性。结论:在胃癌组织中高表达的V-ATPases与TGF-β1可能胃癌发生浸润转移的重要生物学标志,检测V-ATPases与TGF-β1对判断胃癌浸润转移具有重要意义。%Objective: To investigate the expression of V-ATPases and TGF-β1 in gastric cancer and their clinical significance. Methods: Immunohistochemistry and western blot were used to detect the expression of V-ATPases and TGF-β1 in gastric cancer specimens, respectively. The correlations among the V-ATPases, TGF-β1 protein, and clinico-pathological parameters of gastric cancer were analyzed. Results: In the assay of immunohistochemistry showed that the positive expression rates of V-ATPases were 75.5%and 28.8%in gastric cancer and the adjacent tissue, respectively. The positive expression rates of TGF-β1 were 77.8 % and 33.3 % in gastric cancer and the adjacent tissue, respectively. Sig-nificant differences (P<0.01) were found between the two aforementioned groups. The correlation analysis indicated that the expression of V-ATPases had a close positive correlation with that of TGF-β1 in gastric cancer (P<0.01). The up-regula-tion of V-ATPases expression was associated with lymph node metastasis, TNM, vascular invasion and differentiation (P<0.01). The expression of the TGF-β1 expression was associated with an advanced clinical stage, infiltrating depth, lymph node metastasis and vascular invasion in the cancer (P<0.01). Conclusion:The high expression of V-ATPases and t TGF-β1 protein may be important biological markers for malignant transformation in the invasion and metastasis of gastric can-cer. The detection of V-ATPases and TGF-β1 expression in gastric cancer is significantly important for improving the pre-diction of invasion and metastasis.
