基于响应面法优化红曲霉菌丝体中红色素的提取工艺

         

摘要

Extraction was applied to the mutant strains of Aspergillus ( monascus rubber YT1 ) fermented mycelia, optimized the extraction process of the red pigment, using single-factor test of the red pigment extracted,the three main factors were investigated (Ethanol concentration, extraction time,extraction temperature).the color value of Red pigment as an index of the three factors Box-Behnken response surface optimization,polynomial quadratic equation prediction model, the optimal value of the equation to solve.The results show that the mycelium of Monascus pigment optimum extraction conditions were: Ethanol concentration 90%, extraction time 100 min,extraction temperature 62 ℃ ,the optimal color values to 799.2(U/g) ,with the forecast error for the 0.467 %.%以突变株红曲霉(Monascus rubber YT1)发酵后菌丝体为原料,优化其红曲色素的提取工艺,采用单因素试验对影响红曲色素提取的三个主要影响因素进行了考察(乙醇体积分数、提取时间、提取温度).以红曲色素的色价为指标,对三因素进行Box-Behnken响应面优化,得到二次多项式回归方程预测模型,对方程进行最优值求解.结果表明菌丝体中红曲色素的最佳提取工艺条件为:乙醇体积分数90%,浸提时间100 min,浸提温度62℃,最优色价值达到799.2(U/g),与预测值误差为0.467%.
