酶解法提取鲜马奶活性肽及其抗氧化活性研究

     

摘要

目的:优选提取鲜马奶抗氧化活性肽的酶解条件。方法以水解度为指标,以酶解温度、酶解时间、加酶量为考察因素,筛选抗氧化活性肽提取工艺参数,测定其还原能力及对羟自由基清除、DPPH、超氧阴离子的清除能力。结果提取鲜马奶中活性肽的最佳酶解条件为温度45℃,酶解时间3 h,加酶量3.5%。其还原能力为96.31%,DPPH清除能力为90.78%,超氧阴离子清除能力为94.28%,羟自由基清除能力为94.56%,与 Vc相比无明显差异。结论鲜马奶中的活性肽具有良好的抗氧化活性,对 DPPH、超氧阴离子、羟自由基均有较好的清除作用。优选得到的酶解工艺稳定、可行,可为抗氧化肽的研究提供理论依据和实验基础。%Objective To optimize enzymatic hydrolysis conditions of the extraction of fresh mare milk an-tioxidant peptide.Methods With the hydrolysis degree as index,hydrolysis temperature,hydrolysis time and amount of enzyme as factors,the extraction processing parameters were screened to determine the re-duction ability of the antioxidant active peptide and the scavenging ability of hydroxyl radical scavenging, DPPH and super anion.Results The optimal enzymatic hydrolysis condition of the extraction of active peptide of fresh mare milk was 45 Celsius,hydrolysis time of 3 h,and the amount of enzyme of 3.5%.The reduction ability was 96.31%,DPPH was 90.78%,super anion 94.28% and the scavenging ability of hy-droxyl radical scavenging was 94.56%,which there were no significant differences compared with that of Vitamin C.Conclusion The bioactive peptides of fresh mare milk possesses the good antioxidant activity, and the good scavenging effect for DPPH, superoxide anion, and hydroxyl radical.The optimized enzymatic hydrolysis process is stable and feasible,which can provide theoretical and experimental basis for the study of antioxidant peptides.
