原发性肺癌组织中 VEGF、HIF-1α、EGFR 的表达及其相关性研究

摘要

Objective To investigate the expression and its correlation of vascular endothelial growth factor (VEGF),hypoxia inducible factor (HIF-1α)and epidermal growth factor receptor (EGFR)in primary lung cancer.Methods 63 cases of primary lung cancer and 26 cases of benign lung disease were collected to detect the expression of VEGF HIF-1α and EGFR with immunohistochemical method in the lung cancer tissues and benign lung lesions.Results The positive rate of VEGF,HIF-1α and EGFR in the primary lung cancer tissues were all significantly higher than that in the lung benign lesions (68.25% vs 20.00%,P<0.001;55.56% vs 13.33%,P <0.001;47.62% vs 13.33%,P =0.001 respectively).The positive ex-pression of VEGF vs HIF-1α,VEGF vs EGFR and HIF-1α vs VEGF in the primary lung cancer tissues were positively correlated (r =0.279,0.295 and 0.267 respectively,P <0.05 for the three couples).The positive expression of and HIF-1αin the primary lung cancer tissues was correlated with lymph node me-tastasis,smoking history and clinical stage,and the positive expression of EGFR was related to lymph node metastasis and clinical stage.Conclusion VEGF,HIF-1αand EGFR were involved in the occurrence and development of primary lung cancer,and there was a synergistic effect among the three.%目的：探讨原发性肺癌中血管内皮生长因子（VEGF）、低氧诱导因子（HIF-1α）、表皮生长因子受体（EGFR）的表达及其相关性。方法收集新疆医科大学附属肿瘤医院手术切除的63例原发性肺癌和26例肺良性病变标本，采用免疫组织化学方法检测原发性肺癌组织和肺良性病变组织中 VEGF、HIF-1α、EGFR 的表达。结果原发性肺癌组织中 VEGF 阳性表达率为68．25％，显著高于肺良性病变组织的20．00％，差异有统计学意义（P ＜0．001）；原发性肺癌组织中 HIF-1α阳性表达率为55．56％，显著高于肺良性病变组织的13．33％，差异有统计学意义（P ＜0．001）；原发性肺癌组织中 EGFR 阳性表达率为47．62％，高于肺良性病变组织的13．33％，差异有统计学意义（P ＝0．001）。原发性肺癌组织中 VEGF 和 HIF-1α的阳性表达呈正相关（r ＝0．279，P ＜0．05）；VEGF与 EGFR 的阳性表达呈正相关（r ＝0．295，P ＜0．05）；EGFR 与 HIF-1α的阳性表达呈正相关（r ＝0．267，P ＜0．05）。原发性肺癌组织中 VEGF、HIF-1α的阳性表达与淋巴结转移、吸烟史和临床分期相关；EGFR 的阳性表达与淋巴结转移和临床分期相关。结论VEGF、HIF-1α、EGFR 在原发性肺癌的发生、发展发挥作用，且可能存在协同作用。
