芝麻油中芝麻素提取与纯化工艺研究

             

摘要

Alcohol as extraction solvent, the optimum condition of extraction of sesamin from sesame oil was obtained by single factor tests and orthogonal test. Then the crude extract sesamin was purified with alcohol solvent, determining the sesamin melting temperature and analyzing by HPLC. The result showed that the optimum conditions for the extraction are as followings: solid- liquid ratio 1 : 5, extraction time 3 h, temperature 50 ℃, extracting times 3. The crude extract accounted for 0. 336 g/100 g from sesamin oil. The crude extract was further purified by crystallization technology. The melting range of the purified sesamin is 120. 5-121.7 ℃. The sesamin content of the purified sesamin reaches 95.2% by HPLC analysis. High purified sesamin was prepared by this work. It would he help to developing high additional value sesamin products.%以芝麻油为原料,用乙醇作提取剂,通过单因素试验和正交试验确定芝麻素最佳提取条件。采用乙醇对芝麻素粗品进行结晶纯化,测定其熔点,并利用高效液相色谱法测定其纯度。结果表明料液比为1:5,浸提时间3h,温度50℃,浸提3次,芝麻素提取率最高,达到0.336g/100g芝麻油。芝麻素结晶熔点为120.5~121.7℃,HPLC法分析芝麻素物质相对含量达95.2%。本研究得到较高纯度的芝麻素纯化结晶,为开发高附加值的芝麻素产品奠定了良好的基础。
