UbcH10在肝细胞癌中的表达及临床病理意义

摘要

Objective To investigate UbcHIO expression in hepatocellular carcinoma and explore its clinicopathological implications. Methods We detected UbcHIO mRNA expression using RT-PCR in normal liver cell line, cancer cell lines, surgically removed hepatocellular carcinoma tissue and corresponding adjacent non-tumor tissue and evaluated the clinicopathological significance of UbcHIO. Immunohistochemistry was performed to investigate UbcHIO protein expression in hepatocellular carcinoma tissue, the adjacent tissue, and normal liver tissue specimens. Results Normal liver cell line L02 showed significantly lower UbcHIO mRNA expression levels than the cancer cell lines BEL-7402, Hep3B, HepG2 and SMMC-7721 (P<0.05). UbcHIO mRNA expression was also was significantly higher in hepatocellular carcinoma tissues than in the corresponding non-tumor tissues (PO.05). Clinicopathological evaluation suggested that UbcHIO expression was associated with tumor invasion of the portal vein, tumor size, TNM staging, and tumor differentiation (P0.05). Immunohistochemistry identified stronger UbcHIO expression in hepatocellular carcinoma tissues than in the adjacent tissues and normal liver tissues (68.6%, 28.6%, and 26.7%, respectively). Conclusion UbcHIO is over-expressed in hepatocellular carcinoma and may serve as a novel biomarker as well as a therapeutic target of hepatocellular carcinoma.%目的 研究UbcH10在肝细胞癌中的表达及其与临床病理特征的联系,初步探讨UbcH10在人肝细胞癌中的生物学意义.方法 利用RT-PCR法检测正常肝脏细胞系及肝癌细胞系,和肝癌组织及对应的癌旁组织中UbcH10 mRNA的表达水平.分析UbcH10 mRNA的表达水平与肝癌临床病理特征之间的关系.利用免疫组化方法检测UbcH10蛋白在肝癌组织及其癌旁组织和正常肝脏组织中的表达.结果 正常肝脏细胞系L02中UbcH10 mRNA表达水平显著低于肝癌细胞细胞系BEL-7402、Hep3B、HepG2、SMMC-7721中UbcH10 mRNA的表达水平(P＜0.05).肝癌组织中UbcH10 mRNA的表达水平显著高于其癌旁组织(P＜0.05).UbcH10mRNA表达水平与肿瘤直径、组织病理分化程度、TNM分期、门静脉侵犯特临床病理特征显著相关(P＜0.05).肝癌组织中广泛表达UbcH10蛋白,总体阳性率为68.6%(48/70),癌旁组织和正常肝组织中较少见UbcH10蛋白阳性表达,其染色阳性率分别28.6%(20/70)和26.7%(8/30).结论 UbcH10在肝癌细胞系及肝癌组织中高表达.UbcH10的高表达与肝癌的恶性临床病理特征相关,UbcH10可能成为潜在的分子标志物或治疗靶点.
