基于UPLC/Q-TOF MS代谢组学技术分析影响远志商品药材质量的关键因素

         

摘要

目的:针对性地分析了产地、生长方式、药用部位等因素对山西产远志商品药材质量的影响,并找出关键影响因素。方法首先将样品进行超高效液相色谱-四极杆-飞行时间质谱分析,并采用主成分分析及聚类分析等多元统计学方法,分别比较了产地、生长方式、药用部位等因素对山西产远志商品药材质量的影响。结果①产地因素是影响现有远志商品药材质量的关键因素,指标性成分3,6’-二芥子酰基蔗糖与远志口山酮Ⅲ的含量高低可以间接反映出远志药材整体质量。②山西石楼、静乐、盂县的野生远志质量较优,而襄汾、闻喜的栽培远志质量较好。③远志筒中化合物的相对含量高于根,根与筒间的主要差异物质为Sibiricoses A6,3,6’-二芥子酰基蔗糖,Tenuifoliose C,远志口山酮Ⅲ,远志根与筒的选择依据应充分考虑临床所治疗疾病的特性,而不能一概而论。结论产地因素是影响现有远志商品药材质量的关键因素,本研究为今后指导远志规范化种植以及产地粗加工和临床精确用药提供了科学依据。%Objective To explore the effect of regions, growth patterns, and medical parts on the quality of commercial Polygala tenuifolia and analyze the key influential factors. Methods The samples of commercial P. tenuifolia were analyzed by the ultra-per-formance liquid chromatography(UPLC)-quadrupole time-of-flight mass spectrometry(Q-TOF MS), and the obtained data were ana-lyzed using principal component analysis, cluster analysis, and other statistical analysis methods. Results The results showed that the key factor affecting the quality of commercial P. tenuifolia was the region, and the contents of 3,6’-disinapoyl sucrose and polygal-axanthone Ⅲ reflected indirectly the whole quality of P. tenuifolia. The quality of wild P. tenuifolia in Shilou, Jingle, and Yu county and cultivated P. tenuifolia in Xiangfen, Wenxi in Shanxi province was better than that from other regions. The relative content of main compounds in roots was higher than in cortices of P. tenuifolia, and the main chemical difference between the roots and cortices were Sibiricoses A6, 3,6’-disinapoyl sucrose, tenuifoliose C and polygalaxanthoneⅢ. Moreover, the choice between the roots and the cor-tices was in accordance with the characteristics of the clinical diseases. Conclusion The key factor affecting the quality of commer-cial P. tenuifolia is the region and this study provides some scientific evidences for the directions of standardized cultivation, habitat processing, and accurate clinical medication of P. tenuifolia in the future.
