响应曲面优化紫花地丁总黄酮提取工艺研究

             

摘要

Optimization of extraction technology of flavonoids from Viola yedoensis Makino by Box-Behnken center composite design.Based on one-factor-at-a-time experiments,the quadratic regression model for the extraction efficiency of total flavonoids was established about the independent variables (ethanol volume fraction,ultrasonic power,the extracting temperature and the extracting time) through Box-Behnken center composite design and response surface methodology.The optimum extraction conditions were as follow:ethanol volume fraction was 64%,ultrasonic power was 161 W,the extracting temperature was 72 ℃,the extracting time was 32 min.Under these conditions,the practical yield of total flavonoids was 4.11%,which was consistent with the predicted yield of total flavonoids of 4.09%.Extraction model of total flavonoids from Viola yedoensis Makino established by Box-Behnken method can precisely predict the experiment results.In the best conditions deduced from the mode,the extraction rate of total flavonoids is high.%运用响应曲面设计(Box-Behnken设计)优化紫花地丁总黄酮提取工艺.在单因素试验基础上,选择乙醇体积分数、超声波功率、提取温度及提取时间为考查因素,采用响应曲面优化超声提取工艺条件,模拟得到总黄酮提取率二次回归方程预测模型.结果表明,最优提取工艺为乙醇体积分数64%、超声波功率161 W、提取温度72℃、提取时间32 min,总黄酮提取率实测结果(4.09%)与响应曲面拟合所得方程预测值(4.11%)符合良好.结果显示,采用Box-Behnken法建立紫花地丁总黄酮提取工艺模型得率高,并能很好地预测试验结果.
