siRNA沉默转录因子OCT1对胃癌细胞株生物学功能的影响

         

摘要

目的 探究沉默转录因子OCT1对胃癌细胞株MGC803和SGC7901生物学功能的影响.方法 应用腺病毒做载体包装OCT1-siRNA,并转染人胃癌细胞株MGC803和SGC7901.应用MTT法和细胞侵袭实验检测 OCT1-siRNA转染是否成功、胃癌细胞增殖和侵袭力的变化.采用Western blot法检测对照组、OCT1-siRNA组及阴性OCT1-siRNA组中人胃癌细胞中OCT1蛋白的表达量.结果 胃癌细胞株MGC803和SGC7901转染OCT1-siRNA 48和72h后,对照组和OCT1-siRNA阴性组细胞增殖能力和细胞侵袭力高/强于OCT1-siRNA组(P均<0.05).MGC803和 SGC7901细胞 OCT1-siRNA组中OCT1蛋白表达水平均下降(F=118.72、91.23, P均<0.05).结论 应用siRNA技术干扰沉默转录因子OCT1的功能后,胃癌细胞株的增殖能力和侵袭力减弱,表明转录因子OCT1可能增强了胃癌细胞的生物学活性.%Objective By using of siRNA to target OCT1 (octamer transcription factor 1) to observe its influence on the biological functions of human gastric cancer cell line MGC803 and SGC7901. Methods Adenovirus mediated OCT1-siRNA was transfected into human gastric cancer cell lines MGC803 and SGC7901. Proliferation and invasion in MGC803 and SGC7901 cells were measured respectively by MTT assay and cells invasion assay after transfection of OCTl-siRNA.The experssion of OCT1 protein in human gastric cancer cell lines MGC803 and SGC7901 was detected after transfection by Western blot. Results After transfection of OCT1-siRNA of 48h and 72h into gastric cancer cell lines MGC803 and SGC7901, the proliferation and invasive ability of the control group and negative OCT1-siRNA group was higher/stronger than that of the OCT1-siRNA group(P<0.05). The OCT1 protein in human gastric cancer cell lines MGC803 and SGC7901 was decresed than control and negative OCT1-siRNA group(P<0.05). Conclusion Using siRNA to target OCT1 function increased proliferation and invasion.lt indicated OCT1 might inhibit biological functions in the gastric cancer cell lines.
