胃癌hMLH1基因启动子CpG岛甲基化

         

摘要

目的 采用特异性甲基化聚合酶链式反应方法检测hMLH1基因启动子CpG岛甲基化水平.方法 对2株胃癌细胞株31例胃癌组织及23例癌旁组织的hMLH1基因启动子CpG岛甲基化水平进行检测.结果 对照组织检测到CpG岛甲基化,胃癌细胞株均检测到CpG岛甲基化,11例胃癌组织检测到CpG岛甲基化,7例癌旁组织检测到CpG岛甲基化.胃癌组织hMLH1基因CpG岛甲基化与分化程度及淋巴结转移之间无相关关系.结论 胃癌存在hMLH1基因启动子CpG岛甲基化,并且可能是胃癌发生的早期分子事件.%Objective To determine the Hypermethylation of CpG island at the hMLHl gene promoter by utilizing methylation-specific polymerase chain reaction analysis. Methods The methylation stata of the hMLHl promoter CpG-island were investigated in 2 human gastric cancer cell lines, 31 primary human gastric carcinomas, and 23 adjacent gastric tissues. Results CpG-island hypermethylaion was not detected in control biopsy specimens but identified in human gastric cancer cell lines and 11 of gastric carcinoma patients. CpG-island hypermethylaion of hMLHl gene was detected in 7 adjacent gastric tissues. In primary gastric tumors, a significant association was not found between tumor differentiation status and lymph node metastasis and CpG island methylation status of hMLHl gene. Conclusion Hypermethylation of CpG island at the hMLHl gene promoter is frequently detected in gastric carcinoma. hMLHl promoter hypermethylation may involve in the early gastric carcinogeneais process.
