摘要:Objective To establish a method for the determination of phenobarbitone (PB), phenytoin(PHT) and carbamazepine (CBZ) in human plasma by reversed-phase HPLC at the same condition.Methods Agilent Zorbax 8 nm Extend-C18 (5 μm, 4.6 mm× 150 mm) was used as analytical column and mobile phase. The UV wavelength was 230 nm and flow rate was 0.8 mL/min. Results PB, PHT and CBZ were separated well. Resolving powers were all more than 1.5. The lowest detective concentrations of PB, PHT and CBZ were as following: 0.3802, 0.1823 and 0.1444μg/mL. The linear ranges were in the range of 0.4~60, 0.2~30, 0.2~30 μg/mL, and relative recovery rates were 97.04%, 96.52% and 97.09%respectively. The relative standard deviations of within-day and between-day were all less than 5.8%.Conclusion The method is simple, sensitive, convenient and reliable. It is suitable for the determination of phenobarbitone, phenytoin and carbamazepine in human plasma at clinic as normal test.%目的 采用反相高效液相色谱法同时测定苯巴比妥(PB)、苯妥英(PHT)和卡马西平(CBZ)血药浓度.方法 色谱柱为Agilent Zorbax 8 nm Extend-C18(5 μm,4.6 mm×150 mm),流动相为V(甲醇)∶V[V(0.1 mol/L乙酸钠)∶V(0.05% β-环糊精)=32∶18]=68∶32,柱温为室温,流速为0.8 mL/min,检测波长为230 nm,进样量为20 μL.结果 PB、DPH和CBZ分离良好,分离度皆大于1.5;最低检测浓度分别为0.3802、0.1823和0.1444μg/mL;线性范围分别为0.4~60、0.2~30和0.2~30 μg/mL;回收率分别为97.04%、96.52%和97.09%;日内和日间相对标准偏差皆小于5.80%.结论 本方法简便、快速、重现性好,结果可靠,适用于临床常规检查.
