大豆苷元对人乳牙牙髓干细胞增殖和成骨分化的影响

         

摘要

目的 探讨大豆苷元对人乳牙牙髓干细胞(SHED)增殖和分化的影响.方法 对体外培养的第3代SHED用浓度分别为0.1、1、10、100 μM的大豆苷元培养基进行培养,采用MTT法检测细胞生长曲线,通过碱性磷酸酶(ALP)试剂盒、放免法和TGF-β1 Elisa试剂盒检测大豆苷元对SHED碱性磷酸酶(ALP)活性、骨钙素(OCN)以及TGF-β1表达的影响.结果 0.1 μM的大豆苷元能够显著促进SHED的增殖,而高浓度(100μM)的大豆苷元则对SHED的增殖具有明显的抑制作用.大豆苷元剂量依赖性地促进了SHED ALP、OCN和TGF-β1的表达.结论 大豆苷元对SHED的增殖和成骨分化具有促进作用,其促成骨分化机制可能与增加TGF-β1表达有关.%Objective To study the effects of daidzein on proliferation and osteogenic differentiation of stem cells derived from human exfoliated deciduous teeth ( SHED). Methods The third passage SHEDs were incubated with 0.1, 1, 10, 100 |xM daidzein, then cells proliferation curves were examined by MTT method. Alkaline phosphatase (ALP) , osteocalcin (OCN) and TGF- (3 1 were detected by alkaline phosphatase kit, radio-immunity method and TGF- f$ 1 Elisa kit, respectively. Results 0.1 (xM daidzein improved proliferation of SHED, 100 (xM daidzein, however, showed a depressant effect on proliferation. ALP, OCN and TGF-|5 1 expression were enhanced by daidzein in a dose-dependent manner. Conclusion Daidzein enhances proliferation and osteogenic differentiation of SHED, and the mechanism may be related with upregulating TGF?{$ 1 expression.
