不同碳源对重组毕赤酵母表达嗜热β-半乳糖苷酶的影响

         

摘要

To analyze the thermostabiliuy of recombinant β - Galactosidase secretory expression level in the constitutive recombinant Pichia pastoris ,glucose ,fructose - glycerol ,lactose ,sucrose and maltose were used as carbon sources respectively . The results showed that monosaccharide and glycerol were better than disaccharide in cell growth and secretory expression of recombinant β -Galactosidase .The secretory expression level of thermostability recombinant β -Galactosidase in fructose was best than the other 5 carbon resources ,glucose second and followed by glycerol ,sucrose better than maltose ,lactose was the lowest .%为分析碳源对嗜热β-半乳糖苷酶在组成型重组毕赤酵母中的表达水平,葡萄糖、果糖、丙三醇、乳糖、蔗糖和麦芽糖分别用于配制培养基的碳源。当以单糖和甘油为碳源时,酵母的生长和重组嗜热β-半乳糖苷酶的分泌水平要优于双糖;以果糖为碳源时,重组酵母分泌嗜热β-半乳糖苷酶的水平最高,其次为葡萄糖和丙三醇,蔗糖要高于麦芽糖,乳糖最低。
