内质网应激蛋白GRP78在宫颈癌组织中的表达及意义

         

摘要

目的:观察内质网应激蛋白GRP78在宫颈癌组织中的表达情况,探讨宫颈癌组织中HPV感染状态与GRP78表达的关系.方法:收集手术切除的宫颈癌组织标本89例,分析高危型HPV感染和低危型HPV感染与患者年龄、病理分化、临床分期及淋巴转移的关系,用免疫组化法检测GRP78在不同HPV亚型感染的宫颈癌组织中的表达情况.结果:在宫颈癌组织中,HPV(+)感染率在低分化鳞癌中高于高分化鳞癌(P<0.05),宫颈癌Ⅱ期中高于宫颈癌Ⅰ期(P<0.05),有淋巴结转移的患者中高于无淋巴结转移的患者(P <0.05);GRP78蛋白在高危型HPV感染的宫颈癌组织中阳性表达率显著高于低危型HPV感染及HPV阴性的宫颈癌组织(P<0.01),而GRP78在低危型HPV感染及HPV阴性的宫颈癌组织中表达无明显差异(P>0.05).结论:HPV(+)感染率随宫颈癌恶性程度增高而增加,高危型HPV感染的宫颈癌组织中GRP78蛋白呈高表达.%Objective:To observe expression of endoplasmic reticulum stress protein GRP78 in cervical cancers and discuss the possible relationship between HPV infection status and GRP78 in cervical cancers.Methods:89 cervical cancer tissue specimens were obtained from surgical resection in department of Obstetrics and Gynecology,and the statistical method was also adopted to analyze the correlations between HPV infection status in cervical cancer tissues and patients' indexes involving age,pathological differentiation,clinical stage and lymph node metastasis,respectively.Then,the expression of GRP78 in cervical cancer tissues infected with different HPV subtypes was detected by immunohistochemical method.Results:In clinical cervical cancer tissues,the infection rate of HPV (+) in cervical cancer tissues were higher in poorly differentiated squamous carcinoma than those in high differentiated squamous carcinoma (P < 0.05).The infection rate of HPV (+) in stage Ⅱ cervical cancer tissues were higher than in stage Ⅰ cervical cancer tissues (P < 0.05).The infection rate of HPV (+) in cervical cancer tissues with lymph node metastasis were higher than those without lymph node metastasis in cervical cancer specimens (P < 0.05).Immunohistochemistry showed that the positive expression rate of GRP78 in high-risk HPV-infected cervical cancer tissues was significantly higher than that in low-risk HPV-infected and HPV-negative cervical cancer tissues (P <0.01),and the expressions of GRP78 in low-risk HPV-infected and HPV-negative cervical cancer tissues had no significant difference (P > 0.05).Conclusions:HPV (+) infection was positively correlated with the degree of malignancy of cervical cancer.GRP78 protein was related to the high-risk HPV infection.
