苦参素对大鼠脑缺血后脑线粒体损伤保护作用的研究

             

摘要

Objective:To study the protective effect of oxymatrine on brain mitochondrial damage after cerebral ischemia.Methods:The model of focal cerebral ischemic was established with Zea-Longa occluding suture,setting sham operating group,the model group and oxymatrine low,medium,high-dose (25,50,100 mg/kg) groups,n =20.10 minutes before operation,the drugs were given by intraperitoneal injection.24 hours later,the extent of mitochondria swelling,membrane fluidity of mitochondria and breathing function of mitochondria in the ischemic brain were measured;the ultrastructural changes of mitochondria by electron microscope was observation;the activity of SOD,GSH-Px,Na+-K+-ATPase,Ca2+-Mg2+-ATPase and the content of MDA were measured;the content of free Ca2+ was measured.Results:Compared with the model group,oxymatrine pretreatment with middle and high dose could obviously improve the mitochondrial swelling,improve the membrane fluidity,improve the mitochondrial respiratory function,inhibit the mitochondrial uhrastructural damage,improve the activity of SOD and GSH-Px and reduce the content of MDA(P<0.05 or P<0.01),and improve the activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase significantly(P< 0.05 or P< 0.01),and decrease the concentration of free Ca2+(P<0.05).Conclusion:Oxvmatrine has a protective effect on brain mitochondrial damage after cerebral ischemia,whose mechanism may be related to its effects of reducing calcium concentration,inhibiting oxidative stress damage and improving ATPase activity.%目的 研究苦参素对大鼠脑缺血后脑线粒体的保护作用.方法 采用线栓法制备局灶性脑缺血再灌注大鼠模型,设假手术组、模型组和苦参素低(25 mg/kg)、中(50 mg/kg)、高(100mg/kg)剂量组,各20只,分别于术前10 min腹腔注射给药.24 h后,测定缺血侧脑线粒体肿胀程度、膜流动性、呼吸功能,电子显微镜观察线粒体超微结构变化,测定线粒体内超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性,MDA含量,Na+-K+-ATPase 、Ca2+-Mg2+-ATPase活性,游离Ca2+浓度.结果 与模型组比较,经苦参素中、高剂量预处理能够明显改善大鼠脑缺血后线粒体肿胀,提高膜流动性,改善线粒体呼吸功能,抑制线粒体超微结构损伤,显著提高SOD 、GSH-Px活性并降低MDA含量(P<0.05或P<0.01),显著提高Na+-K+-ATPase 、Ca2+-Mg2+-ATPase活性(P<0.05或P<0.01),降低游离Ca2+浓度(P<0.05).结论 苦参素对脑缺血后脑线粒体损伤具有一定的保护作用,其机制可能与降低Ca2+浓度、抑制氧化应激损伤以及改善ATPase活性有关.
