植物甾醇-共轭亚油酸酯对结肠癌细胞增殖的抑制作用

The aim of this study is to explore the effects of phytosterin-conjugated linoleic acid on cell proliferation in human colon cancer HT-29 cells and the likely underlying anti-cancer mechanisms.Human colon cancer HT-29 cells were cultured and treated with different concentrations of PCLA (100, 150, 200, 250, 300 μmol/L) and the influence on proliferation was observed. Cell proliferation condition was observed by MTT. Cell morphology character was compared by inverted fluorescence microscope. The condition of proliferation in PCLA groups decreased compared with the control group, there were dose-effect(100 to 300 μmol/L) and time-effect(24, 48, 72 h) relation in different PCLA groups. PCLA can reduce human HT-29 colon cancer cell proliferation.%观察植物甾醇-共轭亚油酸酯对结肠癌HT-29细胞增殖的作用,探讨其可能的作用机制.采用体外培养结肠癌HT-29细胞的方法,施加不同浓度植物甾醇-共轭亚油酸酯(100、150、200、250、300μmol/L),观察对细胞增殖的影响.应用四唑盐比色法(MTT法)观察细胞增殖抑制情况,倒置荧光显微镜观察细胞形态学特征.结果显示,植物甾醇-共轭亚油酸酯各组较对照组增殖降低,且存在剂量-效应关系(100～300 μmol/L)和时间-效应关系(24、48、72 h),说明植物甾醇-共轭亚油酸酯抑制结肠癌HT-29细胞增殖.
