十全大补汤对小鼠骨髓源性细胞向脑内迁移的影响

     

摘要

目的 探讨十全大补汤对小鼠骨髓源性细胞向脑内迁移的影响.方法 将增强型绿色荧光蛋白转基因小鼠的骨髓细胞通过股静脉移植入实验小鼠制作成嵌合鼠模型,4周后分为实验组(口服十全大补汤组)和对照组(给水组),继续饲养5周.利用流式细胞术检测鼠血中GFP及CD1 1b阳性细胞比例,利用免疫组织化学法检测鼠脑GFP或Iba1阳性细胞的分布.结果 骨髓移植成功,嵌合鼠79%以上血白细胞由移植来源的骨髓细胞产生,十全大补汤能够促进血中单核巨嗜细胞系统的增殖.2组小鼠小脑内均偶可见GFP阳性细胞,差异无统计学意义.十全大补汤能够增加小鼠脑海马区固有小胶质细胞的数量.结论 对于无脑部损伤的小鼠,口服十全大补汤能够促进血中CD11b阳性的单核巨噬细胞系统的增殖,同时能够增加脑中固有小胶质细胞的数量.%Objective To investigate the effect of Shiquandabutang on the migration of mouse bone marrow-derived cells into brain. Methods We made chimeric mice model by transplanting the flesh bone marrow cells from enhanced green fluorescent protein transgenic mice into mice through femoral veins. After regular feeding for 4 weeks, experimental group (Shiquandabutang group) and control group (water group) were divided into, then 5 weeks regular feeding continued. The blood GFP and CDllb positive cells were determined by flow cy-tometry. The brain GFP or Ibal positive cells were detected by immunohistochemishy. Results Bone marrow transplantation succeeded, more than 79% of white blood cells in chimeric mice were derived from the transplanted bone marrow cells. Shiquandabutang promoted the proliferation of blood mononuclear macrophage system. Few GFP-positive cells were detected in the cerebellum of two groups, and there was no significant difference. Shiquandabutang increased the inherent microglia within hippocampus. Conclusion In normal mice brain, Shiquandabutang could promote the proliferation of blood CD1 lb-positive monocyte-macrophage system, as well as brain inherent microglia.
